Openlab Application – Colocalization

The Openlab Colocalization module provides rapid visualization and quantitation of colocalization in samples labeled with multiple fluorochromes. For any pair of selected images, the module creates a scatter plot display of intensity distribution and degree of colocalization. Statistical analysis provides both whole image statistics and user defined ROI statistics, including Pearson's Correlation and Colocalization Coefficients.

The degree of colocalization can be displayed and saved as a binary mask for morphological analysis using the Advanced Measurements module. Statistical data can be exported to a spreadsheet for analysis and scatter plot pictures can be exported to use in presentations.

Openlab Application - Registration

This module allows you to correct for any X,Y image displacement which can arise when capturing images at different wavelengths. The module provides an interactive preview window that allows sub-pixel movement of images for precise alignment of multi-wavelength image sequences. Settings can be stored and used again to save time and increase accuracy. Use to correct image alignment before analysis using the Colocalization or FRET modules.

Colocalization case study

The laboratory of Dr George Bloom, Department of Biology and Cell Biology, University of Virginia, is focused on the role of microtubules and microtubule motor proteins in membrane trafficking. Of particular interest is the regulation of microtubule associated proteins in relation to Alzheimer's disease.

The images to the left are the the result of an experiment to compare the localization of the microtubule associated protein, tau, in fibroblasts tranfected with wild type and mutant tau protein. An Openlab cell imaging system was used to acquire images of the two cell types labeled with different fluorochromes. The amount of colocalization of tau in the two cells was then analyzed using the Openlab Colocalization module. The scatter plot displays the intensity distribution and degree of colocalization. The module also provides statistical analysis of the data which can be exported to a spreadsheet.
