Openlab Application – Morphological Measurements

The Openlab Morphology Module Bundle provides a range of tools for morphological analysis and consists of three Openlab modules. The Density Slicing module detects objects in monochrome and color images by density slicing or thresholding and allows the creation of binary images. The Advanced Measurements module upgrades the standard Measurements module with features including automatic measurement of multiple objects in binary images. The Binary Operations module allows Boolean logic to be applied to binary images.

Openlab images can be imported directly into Volocity Quantitation for further and more complex measurement analysis.

Morphological Measurements case study

Dr David Grainger is the head of the Inflammation Research and Therapy Laboratory, University of Cambridge. The group primarily researches into the role of inflammation in cardiovascular disease, with particular focus on cytokines and signalling molecules. Dr Grainger uses an Openlab cell imaging system to measure and quantify various cell types and tissues, such as the mouse liver macrophage cells shown here.

In the left image, the Density Slicing module has been used to create a binary mask to select all the DAPI stained cell nuclei. Using Advanced Measurements, morphological measurements have been made using the binary mask, including area, perimeter, width, height and intensity measurements. The measurements table is saved with the image document and the data can be exported for statistical analysis.
